KNOW Bio Announces Breakthrough Nitric Oxide Discovery to Combat COVID-19

First definitive demonstration of nitric oxide efficacy against the SARS-CoV-2 virus
Research Triangle Park, N.C. (September 10, 2020) – KNOW Bio, LLC has generated the first definitive evidence that nitric oxide (NO) can inhibit viral reproduction of SARS-CoV-2, the human coronavirus that causes COVID-19. Nitric oxide is a molecule produced naturally by the body to regulate blood flow, control inflammation, and defend against invading microorganisms.
These milestone in-vitro experiments were conducted by testing multiple nitric oxide-releasing compounds against wild type SARS-CoV-2 virus at an independent laboratory with a deep history of antiviral research. Therapeutic doses (<200 µM) were applied to infected VERO E6 cells and showed a dose-dependent effect on viral replication. The result was a greater than 99.9% reduction in virus observed after 24 hours versus virus observed on untreated cells.

Furthermore, there was no cytotoxic impact (cell damage or cell death) on uninfected cells.

Previous reports have only speculated on the potential of nitric oxide to serve as a therapeutic for patients with COVID-19 or as a preventative countermeasure for those who are at risk of exposure to the virus.1 These conjectures were based in large part on prior published scientific evidence derived from studies conducted on the original SARS-CoV virus.

Nitric Oxide (NO) has been reported to:
  • Inhibit coronavirus infection by altering spike protein binding and disrupting the ability of the virus to attach to the receptor and infect human cells.2
  • Inhibit coronavirus viral replication inside an infected cell by slowing viral RNA production - resulting in fewer RNA strands and viral copies.3
  • Inhibit release of reproduced virus by preventing host cell death via interfering with caspase activity - the reproduced virus particles thus remain within the infected cell and fewer virus particles are spread to infect other cells.4
“We have gained new understandings into the effective concentrations, rates of release and mechanism of action of nitric oxide against human coronavirus - providing valuable insights into safety and efficacy,” continued Schoenfisch.

Submission for publication and peer review is expected before year end.

Vast Therapeutics Announces BIOC11 testing results for Nontuberculous Mycobacteria (NTM) in Animal Model

Vast's Second Drug Candidate achieves 99% reduction of NTM

DURHAM, N.C., Oct. 22, 2019 -- Vast Therapeutics, Inc., a subsidiary of KNOW Bio, LLC, announces that BIOC11 eradicated 99% of the Nontuberculous Mycobacteria (NTM) in an in vivo animal model of infection; this data will be presented at the National Cystic Fibrosis Conference to be held in Nashville, Tennessee from October 31 to November 2, 2019.

The NTM animal model is broadly recognized by the field as predictive for evaluating efficacy in humans.  Dr. Diane Ordway, whose laboratory at Colorado State University developed the model and conducted the study, noted "To achieve this level of efficacy is impressive, and to do so without any observed negative effects on the animals is particularly exciting." 

"BIOC11 is an important expansion of our pipeline as it looks and acts more like a small molecule drug candidate rather than the larger biopolymers that we have shown efficacy with in the past," said Dr. Mark Schoenfisch, President and CSO of Vast Therapeutics. "Not only did we achieve 99% reduction in NTM counts, the activity was superior to traditional antibiotics, validating our belief in the superiority of our technology over traditional antibiotic treatments being used today. We are equally excited that the use of our drug candidate led to no adverse effects. Our proprietary water-soluble NO-releasing biopolymer technology, which delivers NO safely and at bactericidal levels for extended periods, reflects an advanced platform for the delivery of nitric oxide. The antibacterial activity of BIOC11 in this animal model of infection was similar to that of our other drug candidate, BIOC51, demonstrating that our nitric oxide-releasing biopolymer pipeline is deep with additional candidates. We look forward to progressing our development toward first-in-human clinical trials."

In vitro, BIOC11 has eradicated every bacterial pathogen tested to date, including 25 species of both Gram-positive and Gram-negative bacteria, 21 distinct strains of Pseudomonas aeruginosa, and 35 antibiotic-resistant strains deemed superbugs by the Center of Disease Control and World Health Organization because of their severe threat to public health. Additionally, given the relationship between NTM and Tuberculosis, and the track record of these compounds in eradicating bacterial pathogens, the Company believes these compounds will also be effective against Tuberculosis.

Vast Therapeutics Receives Qualified Infectious Disease Product Status for BIOC11

Second Drug Candidate to Receive QIDP Designation for Vast
DURHAM, NC – October 8, 2019 – Vast Therapeutics, Inc., a subsidiary of KNOW Bio, LLC, today announced that another of its drug candidates, BIOC11, has been designated as a “Qualified Infectious Disease Product” (QIDP) by the U.S. Food and Drug Administration. Vast is a preclinical-stage  pharmaceutical company focused on treating respiratory infections through the power of nitric oxide (NO) in a synthetically derived powder-based form. The QIDP designation, granted for treatment of chronic pulmonary infections due to Pseudomonas aeruginosa in patients with cystic fibrosis, will make BIOC11 eligible to benefit from certain incentives for the development of new antibiotics provided under the Generating Antibiotic Incentives Now Act (GAIN Act). These incentives include priority review and eligibility for fast-track status. Further, if ultimately approved by the FDA, BIOC11 is eligible for an additional five-year extension of market exclusivity.

The QIDP designation was created by the Generating Antibiotic Incentives Now (GAIN) Act and creates incentives for the development of certain antibiotics that treat serious or life-threatening infections. The primary incentives are an additional five-year extension of Hatch-Waxman Act exclusivity, as well as priority FDA review of the first marketing application or efficacy supplement for BIOC11 and the indication for the QIDP designation with an opportunity to request Fast Track designation for BIOC11

KNOW Bio Announces Closing of $30 Million in Equity Financing

Follows $25 Million in Corporate Commitment Last December
​Company’s Novel Anti-Infective Has Robust Runway


Durham, N.C., May 15, 2019 /PRNewswire/ -- KNOW Bio, LLC, (the Company or KNOW Bio), an RTP based conglomerate of biotechnology and MedTech companies, announced today that the Company has completed its next round of financing with a single investor, Reedy Creek Investments, LLC (Reedy Creek).

The financing provides $30 million in additional funds for research & development and general operations. Vast Therapeutics, one of the Company’s majority owned subsidiaries, is working on a range of products with the top objective to deliver solutions to battle respiratory infections in patients with cystic fibrosis (CF). Currently, 80% of the CF population will succumb to infections in what is now an average lifespan of 40 years. The Company has eradicated 29 strains of Pseudomonas aeruginosa (PA), the primary pathogen that causes the majority of exacerbations in CF patients. At all ages, CF patients are at risk of being hospitalized for months at a time using antibiotics capable of only managing the PA infection. The Company’s therapeutics in development have shown efficacy in both in vitro lab testing and in animal models including the ability to eradicate the aggressive bacteria from the sputum of actual CF patients.
